Time resolved sub-cellular singlet oxygen detection – ensemble measurements versus single cell experiments
نویسنده
چکیده
Systematic in vitro studies on singlet oxygen luminescence kinetics have been carried out reconciling recently published contradictory results reported for sub-cellular singlet oxygen kinetics obtained with two different approaches: single cell and cell ensemble measurements. The singlet oxygen luminescence kinetics in two cell lines were investigated after incubation with three different photosensitizers. In each case a strong dependency of the singlet oxygen luminescence kinetics from the applied illumination already at very low doses was observed. Analysis of the obtained results allows judging the usability of the two approaches for detailed investigation of sub-cellular singlet oxygen kinetics. Intrinsically existing detection limits as well as local heating during the measurement will be discussed. 1 O 2 l u m in e s c e n c e , a .u .

Singlet Oxygen Production by PSII Under Light Stress: Mechanism, Detection and the Protective role of β-Carotene
In this review, I outline the indirect evidence for the formation of singlet oxygen ((1)O(2)) obtained from experiments with the isolated PSII reaction center complex. I also review the methods we used to measure singlet oxygen directly, including luminescence at 1,270 nm, both steady state and time resolved. Other methods we used were histidine-catalyzed molecular oxygen uptake (enabling (1)O(...

Photodynamic therapy (PDT) dosimetric tools are crucial for treatment planning and noninvasive monitoring by means of fluorescence. Present approaches consider usually a 1D problem, a simple photochemical process, or a spatially homogeneous photosensitizer. In this work, a radially resolved superficial photosensitizer fluorescence and 3D photochemical time-dependent PDT model are presented. The...
متن کاملA platform for high-throughput bioenergy production phenotype characterization in single cells
Driven by an increasing number of studies demonstrating its relevance to a broad variety of disease states, the bioenergy production phenotype has been widely characterized at the bulk sample level. Its cell-to-cell variability, a key player associated with cancer cell survival and recurrence, however, remains poorly understood due to ensemble averaging of the current approaches. We present a t...
متن کاملSinglet molecular oxygen by direct excitation.
Direct excitation at 1064 nm and detection of singlet molecular oxygen at 1270 nm is made possible by the availability of powerful YAG-lasers and sensitive NIR photomultipliers. Singlet oxygen was generated in condensed phase at 77 K by direct excitation at 1064 nm (without the use of sensitizers). Several luminescing species were observed by time resolved luminescence spectroscopy and luminesc...
متن کاملPhysical mechanisms of generation and deactivation of singlet oxygen.
For more than 70 years, researchers in several areas of science have been intrigued by the physical and chemical properties of the lowest excited states of molecular oxygen. With two singlet states lying close above its triplet ground state, the O2 molecule possesses a very unique configuration, which gives rise to a very rich and easily accessible chemistry, and also to a number of important p...
